山药总蛋白对大鼠睾丸损伤后生精功能的修复作用
Repair of Spermatogenic Function in Rats with Testicular Injury by Yam Total Protein

Abstract
The extraction process of yam total protein was optimized by orthogonal test,and its stabil-ity was verified.The amino acid content in the total protein of yam was determined with an automatic amino acid analyzer.The model of kidney-yang deficiency was established by hydrocortisone in rats,and the effects of yam total protein on the state of rats,testicular index,epididymal index,sperm count and sperm motility,testosterone secretion and the quantity of Leydig cells were observed.At the same time,an oxidative stress model of TM3 cells in vitro by hydrogen peroxide was established to explore reparative effect of yam total protein on testicular function after testicular injury in rats.Results show the optimal extraction process for yam total protein was 20 mmol/L Tris-HCl(pH 7.4)as the extraction solution,the ratio of material to liquid was 1∶10,the leaching was 2 hours,and the ammonium sulfate saturation was 65%.The RSD value of the verification test was 2.89%,which is less than 3%.The average yield of yam total protein was 2.12%.The yam total protein can increase the body weight and organ index of kidney-yang deficiency rats,enhance sperm motility,promote tes-tosterone secretion,increase the content of Leydig cells and the expression of p-ERK/ERK and p-AKT/AKT.Research findings show that the optimized extraction process of yam total protein was stable and feasible,and yam total protein contained a variety of amino acids.Meanwhile,yam total protein can restore the vitality and function of Leydig cells by activating the associated protein of ERK and AKT signaling pathways,further effectively ameliorate testicular function of rats,and re-pair the spermatogenic ability.关键词
