火针对膝骨关节炎大鼠软骨Wnt-4、β-catenin及MMP-13影响
Effects of fire needling on Wnt-4,β-catenin,and MMP-13 in the cartilage of knee osteoarthritis rats

Abstract
Objective To observe the effects of fire needling on the morphology of cartilage tissue and the protein expression of Wnt-4,β-catenin,and matrix metalloproteinase 13(MMP-13)in knee osteoarthritis(KOA)rats.Method Thirty Sprague-Dawley(SD)rats were randomized into a blank control group(n=11)and a modeling group(n=19).Rats in the modeling group received intra-articular injection of 4%papain to establish KOA models.The two groups each randomly contributed 3 rats for model identification.The rest of the rats in the modeling group were randomly divided into a model group and a fire needling group,each consisting of 8 rats.Rats in the fire needling group were intervened by fire needling at bilateral Dubi(ST35)and Neixiyan(EX-LE5),and rats in the blank control and modeling groups did not receive any intervention.The three groups of rats were observed for the body mass growth rate,knee circumference increase rate,and the appearance of the cartilage.The knee cartilage of rats was treated with hematoxylin-eosin(HE)staining and compared using Mankin's score among the three groups.The Western blot was used to determine the protein expression of Wnt-4,β-catenin,and MMP-13.Result Compared to the blank control group after modeling,the model rats had a smaller body mass growth rate(P<0.05),a higher knee circumference increase rate(P<0.05),and a higher Mankin's score(P<0.05),suggesting the success of modeling.After the intervention,the model group showed a smaller body mass growth rate compared to the blank control group(P<0.05),and the fire needling group had a larger body mass growth rate than the model group(P<0.05);compared to the blank control group,the model group had a smaller knee circumference increase rate(P<0.05)and a higher Mankin's score(P<0.05);the fire needling group had a lower Mankin's score than the model group(P<0.05);the relative protein expression levels of Wnt-4,β-catenin,and MMP-13 were higher in the model group than in the blank control group(P<0.05)and were lower in the fire needling group than in the model group(P<0.05).Conclusion Fire needling can inhibit cartilage degeneration and improve KOA symptoms,which may be associated with down-regulating MMP-13 by modulating the Wnt/β-catenin signaling pathway.关键词
