晋南牛ApoA1基因克隆及组织表达谱分析
Cloning and Tissue Expression Profile Analysis of ApoA1 Gene in Jinnan Cattle

Abstract
In order to study the biological structure and function of the apoliprotein A1(ApoA1)gene in Jinnan cattle and its expression in different tissues,the sequence of ApoA1 gene was cloned using total RNA of Jinnan cattle liver as template.Various online analysis softwares were used to analyze the bioinformatics of ApoA1 gene in Jinnan cattle and construct phylogenetic tree.The mRNA expression levels of ApoA1 gene in heart,liver,spleen,lung and kidney of Jin-nan cattle were detected by qPCR.The results showed that the length of CDS region sequence of ApoA1 gene of Jinnan cattle was 852 bp and the homology was 98.00%compared with the ApoA1 gene sequence of Bos Taurus.The molecu-lar weight of ApoA1 protein in Jinnan cattle was 30 276.35 Da,which was composed of 265 amino acid,belonging to hydrophilic protein,and there were 20 phosphorylation sites.The secondary structure of ApoA1 protein belonged to all-alpha protein,in which α-helix accounting for 92.08%.The ApoA1 protein exhibited significant interactions with pro-teins such as ApoA2,ApoB,ApoE,ApoC2,ApoC3,SCARB1,LPL,ABCA1.The phylogenetic tree based on ApoA1 se-quence homology showed that Jinnan cattle had the closest genetic relationship with Bos taurus and the farthest genetic relationship with Bison.The ApoA1 gene was expressed in liver,heart,spleen,lung and kidney of Jinnan cattle with the highest expression in liver and the lowest expression in kidney.关键词
