宁夏地区羔羊源大肠杆菌分离鉴定及病理组织学分析
Isolation,Identification,and Histopathological Analysis of Escherichia coli from Lambs in Ningxia Region

Abstract
This experiment took Escherichia coli(E.coli)from lambs in Ningxia as the research object,aiming to clarify the prevalence of E.coli from lambs,the distribution of virulence genes,and the pathological tissue changes caused by E.coli.From March 2022 to March 2023,a total of 206 samples of lamb tissues and anal swabs with dia-rrhea symptoms were collected from different farms in Ningxia region.Pathogen isolation,identification,and virulence gene testing were performed based on differential culture medium,PCR amplification,and 16S rRNA sequencing.And one target organ confirmed to be infected with E.coli was subjected to histopathological analysis.The results showed that the total detection rate of E.coli from lambs in Ningxia region was 76.2%(157/206).The detection rate was diffe-rent in different regions,with Guyuan city having the highest detection rate of 93.3%(28/30),which was significantly di-fferent from Yinchuan city(51.2%,21/41,P<0.01)and Wuzhong city(80.0%,108/135,P<0.01).The detection rate of di-fferent sampling sites was also different.The detection rate of anal swab was the highest(91.1%,143/157),which was sig-nificantly different from that of heart(30.0%,3/10),lung(33.3%,4/12)and liver(25.0%,3/12,P<0.01).115 strains of E.coli were isolated and identified by Eosin methylene blue and blood plate identification medium.When the virulence factors of different isolates were detected,the detection rates of pilus genes K88 and F14 were 25.2%(29/115)and 8.8%(10/115),the detection rates of virulence island genes HPI and LEE were 84.3%(97/115)and 74.0%(85/115),and the detection rates of toxin genes st1,st2,lt1,stx1 and stx2/stx2e were 23.5%(27/115),40.9%(47/115),2.6%(3/115),55.6%(64/115)and 59.1%(68/115),respectively,and the fimbriae genes K99,987P and F18fedA were not detected in di-fferent isolates.Histopathology showed that villi in the mucosal layer of intestinal tissue were dissolved and necrotic,and the mucosal epithelial structure was incomplete,accompanied by inflammatory cell infiltration such as lymphocytes,neu-trophils and plasma cells.In summary,the infection of pathogenic E.coli in lambs in Ningxia was serious,the detection of pathogenic E.colishould be strengthened,and corresponding prevention and control measures should be taken.关键词
