外泌体介导的circ_0000615通过靶向miR-337-3p调控IL-1β诱导的骨关节炎软骨细胞损伤
Exosome mediated circ_0000615 regulates IL-1β induced chondrocyte injury in osteoarthritis by targeting miR-337-3p

Abstract
Objective:To investigate the effect of circ_0000615 mediated by exosome on IL-1β induced injury of human chon-drocytes CHON-001 and its molecular mechanism.Methods:CHON-001 cells were treated with IL-1β to construct osteoarthritis(OA)model in vitro.The expressions of apoptosis-related proteins were detected by Western blot.RT-qPCR was used to detect the ex-pression levels of circ_0000615 and miR-337-3p.Flow cytometry was used to detect cell apoptosis.The interaction between miR-337-3p and circ_0000615 was analyzed by double luciferase reporter gene assay.The expression of inflammatory cytokines was detected by ELISA.The effect of circ_0000615/miR-337-3p axis on CHON-001 cell damage was observed by functional recovery assay.Results:circ_0000615 and miR-337-3p were up-regulated and down-regulated in OA cartilage tissues and cell models,respectively(P<0.05).Compared with Control group,IL-1β-induced CHON-001 cell apoptosis rate,Bax protein level,IL-6,TNF-α levels were increased,Bcl-2 protein and IL-10 levels were decreased(P<0.05).Exosome-mediated silencing of circ_0000615 decreased apoptosis rate,Bax protein level,IL-6,TNF-α levels,and increased Bcl-2 protein level and IL-10 level of CHON-001 cells(P<0.05).circ_0000615 tar-geted regulation of miR-337-3p expression(P<0.05),and down-regulated miR-337-3p reversed the effect of circ_0000615 silencing on CHON-001 cell damage(P<0.05).Conclusion:Exosome-mediated silencing of circ_0000615 alleviates CHON-001 cell damage through targeted up-regulation of miR-337-3p.关键词
