基于猪流行性腹泻病毒S蛋白单克隆抗体的竞争ELISA方法的建立
Establishment of a competitive ELISA method based on monoclonal antibodies against porcine epidemic diarrhea virus S protein

Abstract
To establish a competitive ELISA method based on porcine epidemic diarrhea virus(PEDV)S protein monoclonal antibody,the main antigen region of PEDV S protein was expressed using an eukaryotic ex-pression system,and monoclonal antibodies against PEDV S protein were prepared after immunizing mice.After optimizing the components and reaction conditions,a competitive ELISA method for detecting PEDV S antibo-dies was established.The S protein was 92 kDa,and an IgG1 subclass anti-S protein monoclonal antibody 5B3D6 was prepared.The established competitive ELISA method had a critical S/N value of 0.400,a short sample detec-tion time(55 min),stable repeatability(coefficient of variation less than 10％),strong specificity(no crossreac-tivity),and a high overall agreement rate with serum neutralization tests(97.08％).This method provides strong support for PEDV antibody evaluation and prevention and control for porcine epidemic diarrhea.关键词
