摘要

Abstract

This aim was to investigate the effects of Ligustrum lucidum polysaccharide(LLP)on the proliferation,apoptosis,secretion,and antioxidant function of TM3 cells exposed to triclocarban(TCC).The cell proliferation activity was determined by CCK-8,and the exposure concentration of TCC as well as the optimal concentration of LLP were screened.The experiment was divided into four groups,control group,LLP group,TCC group,TCC+LLP group.Testosterone secretion levels in TM3 cells were de-termined by ELISA,the mRNA expression levels of genes related to cell proliferation,apoptosis,and testosterone synthase(Bax,Bcl-2,Caspase 3,Caspase 8,StAR)were detected by RT-qPCR.The expression levels of apoptosis-related proteins Bax and Bcl-2 were detected by Western-blot,while SOD activity and MDA content were measured using a reactive oxygen species kit.Additionally,cell reactive oxygen species and apoptosis were detected using flow cytometry.The results showed that when TCC was exposed,cell proliferation activity decreased(P<0.01),testosterone secretion level increased(P<0.01),Bcl-2 mRNA expression level decreased(P<0.05),Bax,Caspase 3,Caspase 8,and StAR mRNA expression levels in-creased(P<0.05).SOD activity decreased(P<0.05),MDA content increased(P<0.01),and apoptosis rate increased(P<0.05).Compared with TCC group,TCC+LLP group significantly increased cell prolifer-ation activity(P<0.01),significantly decreased testosterone secretion level(P<0.01),and signifi-cantly increased Bcl-2 mRNA expression level(P<0.01).The mRNA expression levels of Bax,Caspase 3,Cas-pase 8 and StAR genes were significantly decreased(P<0.05),SOD activity was increased(P<0.01),MDA content,apoptosis rate and ROS levels were decreased(P<0.05).The changes of apoptosis-related pro-teins Bax and Bcl-2 were consistent with the genetic results.These results indicate that LLP can sig-nificantly alleviate the proliferation,apoptosis,abnormal testosterone secretion and oxidative stress of TM3 cells exposed to TCC.
