Network pharmacology and in vitro experimental validation to reveal the mechanism of giant knotweed rhizome against PRRSV

摘要

Abstract

This study aimed to investigate the effects and potential active ingredients and mecha-nisms of Giant knotweed rhizome(GKR)on porcine reproductive and respiratory syndrome virus(PRRSV)based on in vitro experiments and network pharmacology methods.Firstly,the water extract of the GKR was pre-pared,and its cytotoxicity on Marc-145 cells was determined using CCK-8 method,and then the inhibitory effect of the GKR water extract on PRRSV proliferation at different times and doses was determined using RT-qPCR,Western-blot and indirect immunofluorescence assay.Secondly,the active ingredients and corre-sponding targets of the GKR were screened using the TCMSP database and the PRRSV disease targets were screened using the GeneCards database,then their intersection targets were further screened.Furthermore,a protein interaction network was constructed based on the obtained intersection targets,and GO function and KEGG enrichment analysis were performed.Finally,molecular docking validation was performed on the selected main active ingredients and core targets.The results of in vitro experiments showed that the maximum cell safety concentration of the GKR water extract significantly inhibited virus prolifera-tion at 24,36 and 48 hours post PRRSV infection,the high and medium doses of the GKR water extract could inhibit PRRSV proliferation in a dose-dependent manner.The results of network pharmacology showed that there are 11 active ingredients in the GKR,which can act on 212 anti-PRRSV targets.GO func-tional enrichment analysis yielded 1383 entries,while KEGG pathway enrichment analysis yielded 179 signaling pathways.Molecular docking showed that the main active ingredients of the GKR,resveratrol,quercetin,and luteolin,can bind well to the core targets such as interleukin-6(IL-6),serine/threonine kinase(AKT1),tumor necrosis factor(TNF),interleukin-1β(IL-1β),tumor protein P53(TP53),and AP-1 transcription factor subunit(JUN).The research results indicated that the GKR may exert anti-PRRSV effects by acting on targets such as IL-6,AKT1,TNF,IL-1β,P53 and JUN through active ingredients such as resveratrol,quercetin and luteol in.
