目的:观察温中消痞汤对脾胃虚寒型功能性消化不良大鼠胃肠动力及血清胃动素(MTL)、胃泌素(GAS)、血管活性肠肽(VIP)的影响,探讨其治疗脾胃虚寒型功能性消化不良的作用机制.方法:70只6日龄SPF级雌鼠随机分为空白对照组(15只)、模型对照组(15只)、莫沙必利组(10只)和温中消痞汤高、中、低剂量组(各10只).除空白对照组外,其余各组采用1%碘乙酰胺联合2%蔗糖灌胃,然后给予4℃食醋灌胃复制脾胃虚寒型功能性消化不良模型.空白对照组和模型对照组给予生理盐水灌胃,温中消痞汤低、中、高剂量组及莫沙必利组分别给予温中消痞汤(7.1、14.2、28.4 g/kg)和莫沙必利(1.575 mg/kg)灌胃,连续30 d.测定胃排空率及小肠推进率,HE染色观察胃组织病理改变情况,Elisa法检测血清MTL、GAS、VIP的含量.结果:与模型对照组比较,各药物干预组精神状态、活动度、粪便性状、进食量、体质量明显改善,大鼠胃排空率显著升高(P<0.01);血清MTL、GAS升高(P<0.01,P<0.05),莫沙必利组和温中消痞汤高剂量组小肠推进率升高(P<0.01,P<0.05),血清中VIP水平降低(P<0.01),温中消痞汤中剂量组血清中VIP降低(P<0.01).结论:温中消痞汤能够通过提高脾胃虚寒型功能性消化不良大鼠胃排空率、小肠推进率,促进大鼠血清内MTL、GAS表达,抑制VIP表达,从而改善胃肠动力,发挥治疗功能性消化不良的作用.

Objective:To observe the effects of Wenzhong Xiaopi Decoction on gastrointestinal motility and serum levels of motilin(MTL),gastrin(GAS),and vasoactive intestinal peptide(VIP)in rats with functional dyspepsia of spleen-stomach deficiency and cold syndrome,and to explore the mechanism of its therapeutic action for this syndrome.Methods:Seventy 6-day-old SPF female rats were randomly divided into a blank control group(15 rats),model control group(15 rats),mosapride group(10 rats),and Wenzhong Xiaopi Decoction high-,medium-,and low-dose groups(10 rats each).Except for the blank control group,all other groups were modeled for functional dyspepsia of spleen-stomach deficiency and cold syndrome by administering 1%iodoacetamide combined with 2%sucrose,followed by cold(4℃)vinegar gavage.The blank control and model control groups received saline gavage,while the Wenzhong Xiaopi Decoction low-,medium-,and high-dose groups and the mosapride group received Wenzhong Xiaopi Decoction(7.1,14.2,28.4 g/kg)and mosapride(1.575 mg/kg)gavage,respectively,for 30 consecutive days.Gastric emptying rate and small intestine transit rate were measured,gastric tissue pathology was observed via HE staining,and serum MTL,GAS,and VIP levels were measured using ELISA.Results:Compared with the model control group,all drug intervention groups showed significant improvement in mental state,activity levels,stool quality,food intake,and body weight;gastric emptying rate significantly increased(P<0.01);serum MTL and GAS levels increased(P<0.01,P<0.05),while VIP levels in serum decreased(P<0.01),particularly in the mosapride and high-dose Wenzhong Xiaopi Decoction groups(P<0.01,P<0.05).Conclusion:Wenzhong Xiaopi Decoction improves gastrointestinal motility by increasing gastric emptying rate and small intestine transit rate,promoting MTL and GAS expression,and inhibiting VIP expression in rats with functional dyspepsia of spleen-stomach deficiency and cold syndrome,thus showing therapeutic effects on functional dyspepsia.
