目的:研究怀牛膝-何首乌药对不同配比对骨质疏松模型小鼠的治疗效果,确定抗骨质疏松效果的最佳比例.方法:将130只KM小鼠(雌雄各半)随机分为模型组、空白组、阳性药和给药组(怀牛膝∶何首乌=1∶2、2∶1、3∶2、2∶3、1∶1、4∶3、3∶4、5∶2、0∶1、1∶0),每组10只.模型组、阳性药、给药组均灌胃维甲酸复制小鼠骨质疏松模型,空白组灌胃等体积生理盐水,将不同配比怀牛膝-何首乌药对加水回流,灌胃给药,阳性药为阿仑膦酸钠.造模12 h后给药,连续15 d.末次给药24 h后取血,离心取上清液,应用ELISA法检测RANKL、OPG、PTH、TRACP-5b、BALP水平,并计算OPG/RANKL.结果:与空白组比较,模型组小鼠BALP、OPG、OPG/RANKL显著降低(P<0.01,P<0.05),TRACP-5b、RANKL、PTH显著升高(P<0.05,P<0.01),表明小鼠骨质疏松模型复制成功.不同给药组对TRACP-5b、PTH、BALP、OPG、RANKL和OPG/RANKL均有影响,表明怀牛膝-何首乌药对具有促进骨形成和抑制骨吸收的作用.综合评分结果显示,1∶2、2∶1、3∶2、2∶3、1∶1、4∶3、3∶4、5∶2、0∶1、1∶0组分别为4.98、2.77、2.89、2.13、2.65、2.53、3.11、2.60、2.14、1.63分,其中怀牛膝-何首乌药对抗骨质疏松作用效果1∶2组评分最高.结论:怀牛膝、何首乌单味药和怀牛膝-何首乌药对均对骨质疏松有一定的治疗作用,但怀牛膝-何首乌药对较单独使用可显著提高对小鼠骨质疏松的治疗作用,以怀牛膝-何首乌药对1∶2配比效果最佳.

Objective:To investigate the therapeutic effects of different ratios of the Achyranthes Radix et Rhizoma(Huai Niu Xi)and Polygoni Multiflori Radix(He Shou Wu)couplet medicinals on osteoporotic mice and to determine the optimal ratio for anti-osteoporosis efficacy.Methods:A total of 130 KM mice(half male and half female)were randomly divided into a model group,blank group,positive drug group,and treatment groups with different ratios of Achyranthes Radix et Rhizoma and Polygoni Multiflori Radix(1∶2,2∶1,3∶2,2∶3,1∶1,4∶3,3∶4,5∶2,0∶1,1∶0),with 10 mice in each group.The model,positive drug,and treatment groups were administered retinoic acid by gavage to induce osteoporosis,while the blank group received saline.The herbal couplet medicinalss were prepared through water reflux and administered by gavage,with alendronate sodium as the positive drug.Treatment began 12 hours post-modeling and continued for 15 days.Blood was collected 24 hours after the final administration,and ELISA was used to measure RANKL,OPG,PTH,TRACP-5b,and BALP levels,as well as calculate the OPG/RANKL ratio.Results:Compared to the blank group,BALP,OPG,and OPG/RANKL were significantly reduced in the model group(P<0.01,P<0.05),while TRACP-5b,RANKL,and PTH levels were significantly elevated(P<0.05,P<0.01),confirming the successful induction of osteoporosis in the model group.Different treatment groups impacted TRACP-5b,PTH,BALP,OPG,RANKL,and OPG/RANKL levels,indicating that the Achyranthes Radix et Rhizoma and Polygoni Multiflori Radix couplet medicinals promotes bone formation and inhibits bone resorption.Comprehensive scoring results were 4.98,2.77,2.89,2.13,2.65,2.53,3.11,2.60,2.14,and 1.63 for the ratios 1∶2,2∶1,3∶2,2∶3,1∶1,4∶3,3∶4,5∶2,0∶1,and 1∶0,respectively,with the 1∶2 ratio showing the highest anti-osteoporosis efficacy.Conclusion:Both Achyranthes Radix et Rhizoma and Polygoni Multiflori Radix alone and in combination have a therapeutic effect on osteoporosis,but the combination is more effective,with the 1∶2 ratio yielding the best results.
