健脾止动汤对慢性束缚应激抽动障碍大鼠多巴胺系统的影响
Effect of Jianpi Zhidong Decoction on dopamine system in rats with chronic restraint stress tic disorder

Abstract
Objective To investigate the effect of Jianpi Zhidong Decoction on dopamine(DA)system in rats with chronic restraint stress tic disorder(TD).Methods 48 male SD rats were selected and randomly divided into the control group,the model group,the tiapride group and the Jianpi Zhidong Decoction group,with 12 rats in each group.After the intervention,the levels of DA,dihydroxyphenylacetic acid(DOPAC)and homovanillic acid(HVA)in brain tissue were detected by high-performance liquid chromatography(HPLC);Western Blot and RT-PCR were used to detect the levels of dopamine D1 receptor(DRD1)and DRD2 proteins,and mRNA expression in the brain.Results Compared with the blank group,the levels of DA and HVA in the hippocampus,the levels of DA,DOPAC and DRD2 protein,and DRD1 mRNA and DRD2 mRNA expression in the hypothalamus in the model group were significantly increased(P<0.05);Compared with the model group,the levels of DA and DOPAC,and the expression of DRD2 protein in the hypothalamus in the tiapride group were significantly decreased(P<0.05);The levels of DA,DOPAC and HVA in hippocampal tissue,the levels of DA,DOPAC and HVA,and DRD1 protein and DRD1 mRNA expression in hypothalamic tissue were significantly decreased in the Jianpi Zhidong Decoction(P<0.05);Compared with the tiapride group,the HVA content in the hippocampus in the Jianpi Zhidong Decoction group was significantly decreased(P<0.05).Conclusion Jianpi Zhidong Decoction intervention can reduce the levels of DA,DOPAC and HVA in the hippocampus and hypothalamus of TD rats with chronic restraint stress Tic Disorders through DRD1-related targets,and improve the function of the DA system,thereby achieving anti-tic effect.关键词
