小檗碱减轻鱼藤酮诱导的帕金森病模型大鼠损伤的实验研究
Experimental study of berberine alleviating rotenone-induced damage of Parkinson's disease model rats

Abstract
Objectives:To investigate the effects of berberine(BBR)on rotenone-induced Parkinson's disease(PD)model rats and its mechanisms.Methods:36 Healthy male SD rats were randomly assigned into the following four groups:A control group(Con),a Rot group,a Rot+BBR group,and a Con+BBR group.Behavioral assessments were performed in each group,and the changes in dopamine content and oxidative stress indicators were observed.Meanwhile,tyrosine hydroxylase(TH),dopa-mine transporter(DAT),and vesicular monoamine transporter 2(VMAT2)expression levels were detected in each group.Rot+BBR group was further treated with adenosine monophosphate-activated protein kinase(AMPK)inhibitor dorsomorphin(Dor)intervention.12 Rats were randomly assigned into a Rot+BBR group and a Rot+BBR+Dor group.The expression levels of APMK,p-AMPK,LC3-Ⅰ,LC3-Ⅱ,and α-synuclein were detected in the Rot+BBR group and Rot+BBR+Dor groups to de-termine the effects of BBR on AMPK signaling pathway-mediated autophagy.Results:Compared to the control group,the rats in the Rot group manifested neurological dysfunction,oxidative stress injury,decreased dopamine content,and decreased expression levels of TH,DAT,and VMAT2.Compared to the Rot group,the motor function of the rats was significantly improved in the Rot+BBR group after BBR treatment.Moreover,the degree of ifluorescence intensity improvement was associated with the BBR dosage.After 20 mg/kg of BBR treatment,oxidative stress injury was alleviated,and the content of dopamine in striatum brain tis-sue of the rats in the Rot+BBR group was increased.Additionally,the proportions of p-AMPK/AMPK and LC3-Ⅱ/LC3-Ⅰ sig-nificantly reduced in the Rot+BBR group after using AMPK inhibitor Dor.Futhermore,the increased expression of α-synuclein and decreased TH-positive fluorescence intensity were also found in the Rot+BBR+Dor group.Conclusions:BBR can alleviate neurological dysfunctions and oxidative stress damage in Rot-induced PD model rats,and the mechanism may be associated with activating AMPK pathway-mediated autophagy.关键词
