巨噬细胞极化在人翼状胬肉发病中的作用研究
Macrophage polarization regulates the development of human pterygium
摘要
Abstract
Objective:To investigate the role of macrophage polarization in the pathogenesis of human pterygium.Methods:48 Cases(48 eyes)of pterygium specimens that met the inclusion criteria were collected from the ophthalmology department of the Second Affiliated Hospital of South China University from June 2023 to April 2024.Among them,24 cases of stationary pterygi-um and 24 cases of progressive pterygium were collected.Additionally,24 cases of normal conjunctiva obtained from donated eye-balls were selected as the control group.Collect specimens and use hematoxylin-eosin(HE)staining to evaluate pathological mor-phological changes in each group;Enzyme-linked immunosorbent assay(ELISA)for detecting the expression of inflammatory fac-tors(IL-1β,TNF-α,IL-6)and anti-inflammatory factors(IL-10);Real-time fluorescence quantitative PCR(RT-qPCR)was used to detect the relative expression levels of M1 type marker iNOS and M2 type marker Arg-1 RNA;Immunofluorescence staining was used to detect the expression of M1 type marker CD68 and M2 type marker CD206.Results:HE staining showed an increase in the number of epithelial cell layers,accompanied by the formation of a large number of new blood vessels and infiltra-tion of inflammatory cells,with more pronounced progression.ELISA showed that compared with the control group,the expres-sion of inflammatory factors in pterygium group was increased,and the increase was more significant in the advanced group.The expression of anti-inflammatory factors decreased,and the decrease was more obvious in the advanced group(P<0.05).RT-qP-CR detection and immunofluorescence staining showed that compared with the control group,the expression of M1 type polariza-tion markers and M2 type polarization markers in the pterygium group was increased,with M1 type markers mainly increasing in the progression phase and M2 type markers mainly increasing in the quiescence phase(P<0.05).Conclusion:Macrophage polar-ization is involved in the development of human pterygium,and its mechanism may be related to the imbalance of macrophage po-larization and the aggravation of ocular surface inflammation.关键词
