摘要

Abstract

Osteoporosis is a metabolic bone disease characterized by decreased bone mass,increased bone fragility,and sus-ceptibility to fractures.Its occurrence depends on the dynamic balance between osteoblasts and osteoclasts.The inactivation of os-teoblasts is closely related to bone remodeling and the maintenance of bone homeostasis.As a new type of cell death,ferroptosis is mainly caused by iron accumulation and abnormal amino acid metabolism,which leads to an increase in the concentration of reac-tive oxygen species in cells,and then iron-dependent lipid peroxidation,inducing cell ferroptosis.Under oxidative stress,Nrf2,an important antioxidant nuclear transcription factor,can improve the antioxidant capacity of cells by activating downstream ARE-specific elements,thereby interfering with the ferroptosis process in osteoblasts.In this paper,the mechanism of iron-dependent cell death and related pathways mediating ferroptosis in osteoblasts are summarized,so as to broaden the ideas for the prevention and treatment of related diseases caused by bone homeostasis imbalance.
