白杨素通过抑制铁死亡减轻脑出血后神经损伤
Chrysin attenuates nerve damage after intracerebral hemorrhage by inhibiting ferroptosis

Abstract
Objective:To investigate the effect of chrysin on nerve injury after intracerebral hemorrhage in rats with ferroptosis as the drug target.Methods:A total of 66 healthy male SD rats were divided into the Sham group(sham operation group),the ICH group(intracerebral hemorrhage model group)and the ICH+Chrysin group(intracerebral hemorrhage model+Chrysin group),with 22 rats in each group.The autologous tail vein blood was injected into the caudate nucleus to establish a model of in-tracerebral hemorrhage,and the same amount of isotonic saline was used to replace the autologous tail vein blood in the Sham group.Z-longa score was performed 6 h after the model was established,and the ICH+Chrysin group was administered by contin-uous gavage for 3 days,and the same amount of normal saline was given to the Sham group and the ICH group.H&E and Nissl staining and transmission electron microscopy were used to observe the morphological changes of brain tissue and hippocampal neu-rons.The contents of Fe2+,GSH,MDA and SOD were detected by the kits,and the expressions of ACSL4,GPX4,FTL,IL-1β and TNF-α in the hippocampus were detected by Western blotting.Results:(1)Compared with the Sham group,the Z-longa score in the ICH group was significantly increased,the brain tissue and hippocampal neurons were significantly damaged,and the ratio of live cells of neurons in the hippocampal CA1 and CA3 regions was decreased,and the differences between the groups were statistically significant(all P<0.01).Compared with the ICH group,the damage of brain tissue and hippocampal neu-rons in the ICH+Chrysin group was reduced,and the proportion of live neurons in the hippocampus CA1 and CA3 regions was in-creased,and the differences between the groups were statistically significant(all P<0.01).(2)Compared with the Sham group,the expressions of Fe2+,MDA,ACSL4,IL-1β and TNF-α in the ICH group were significantly increased,and the expressions of GSH,SOD,GPX4 and FTL were significantly decreased in the ICH group,and the differences between the groups were statisti-cally significant(all P<0.01).Compared with the ICH group,the levels Fe2+,MDA,ACSL4,IL-1β,TNF-α,GSH,SOD,GPX4 and FTL was reversed,and the differences between groups were statistically significant(all P<0.01).Conclusion:Chrys-in attenuates nerve injury after intracerebral hemorrhage in rats by inhibiting ferroptosis.关键词
