Mechanism of Guizhi Fuling capsule in regulating ovarian cancer cell cycle via Caspase-4/GSDME pathway-mediated pyroptosis

Abstract
Objective:To investigate the effect of Guizhi Fuling capsule on Caspase-4/GSDME pathway-mediated pyroptosis and to elucidate its molecular mechanism in regulating the cell cycle for the treatment of ovarian cancer.Methods:Human ovarian cancer cell line SKOV3 was divided into the blank group,the low,medium,and high dose Guizhi Fuling capsule groups,and the inhibitor group,with corresponding drug interventions.Cell viability was assessed using CCK8 assay;lactate dehydrogenase activ-ity in the cell supernatant was measured by ELISA;cell cycle distribution and pyroptosis levels were analyzed using flow cytome-try;and the expression of proteins related to Caspase-4/GSDME pathway and cell cycle was detected by Western blot.Results:Compared to the blank group,Guizhi Fuling capsule significantly inhibited SKOV3 cell viability,reduced the activity of LDH in the cell supernatant and the percentage of cells in the S phase,while increasing the proportion of cells in the G0/G1 phase,with sta-tistically significant differences(P<0.01).Additionally,Guizhi Fuling capsule significantly downregulated the expression of CDK4,CDK6,Cyclin D1,and the ratio of p-Rb/Rb,cleaved Caspase-4/Caspase-4,and cleaved GSDME/GSDME,as well as decreased the proportion of Caspase-1+PI+SKOV3 cells,with statistically significant differences(P<0.01).Conclusion:Guizhi Fuling capsule exerts its therapeutic effect on ovarian cancer by inhibiting the activation of the Caspase-4/GSDME signaling path-way,reducing pyroptosis levels,and inducing G0/G1 cell cycle arrest.关键词
