摘要

Abstract

Objective:This paper intends to investigate the anti-nasopharyngeal carcinoma effect of Pteris semipinnata L(PsL)and its mechanism through network pharmacology combined with in vitro experiments,which will provide a basis for subse-quent studies.Methods:The main active ingredients of PsL were obtained from literatures,and the genes related to the main in-gredients of PsL were obtained from the PharmMapper database,and the genes of nasopharyngeal carcinoma diseases were ob-tained from GeneCard,OMIM,PharmGkb,TTD and CTD databases.The intersection genes of drugs and diseases were import-ed into the String platform to construct protein-protein interactions(PPI)network,and the core targets were screened through the PPI network.The core targets were analyzed by bioinformatics,and the molecular docking of the screened core targets was per-formed and the docking results with low binding energy were visualized.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analyses were also performed.Nasopharyngeal carcinoma Hone1 cells were acted on using PsL extracts,and cell proliferation was detected by CCK-8 method;cell activity was detected by fluorescent staining with Calcein/PI;apoptosis was detected by flow double staining with Annexin V-FITC,and protein kinase B(AKT),phosphorylated AKT(p-AKT),and non-receptor-type tyrosine protein kinase(SRC)protein expression were detected by using Western blot.Results:A total of 115 genes intersecting with nasopharyngeal carcinoma at PsL were obtained.Among them,SRC,MAPK1,and RHOA may be the key targets for nasopharyngeal carcinoma treatment.Bioinformatics analysis showed that SRC was overex-pressed in nasopharyngeal carcinoma tissues,and molecular docking results showed that β-carotene,rutin and apigen-in-7-O-β-D-glucopyranoside glucoside had low binding energy to SRC.The analysis of enrichment results indicated that PsL could regulate PI3K-AKT and other signaling pathways.In vitro experiments showed that the aqueous extract of PsL could inhibit the proliferation and activity of Hone1 nasopharyngeal carcinoma cells and promote the apoptosis of Hone1 cells.The aqueous extract of PsL significantly down-regulated the expression of p-AKT and SRC proteins.Conclusion:PsL can inhibit the proliferation and promote the apoptosis of nasopharyngeal carcinoma cells by negatively regulating the SRC-AKT signaling pathway.
