Abstract
The brown planthopper(BPH),Nilaparvata lugens,is one of the most serious pest of rice.The present pest control strategies show certain limitations,so it is necessary to research and explore novel methods for controlling BPH.In this study,we used RNA interference(RNAi)technology mediated by double-stranded RNA(dsRNA)with sugar transporter 4(Nlst4)of N.lugens as target gene to synthesize the specific dsRNA of Nlst4 in vitro.The transcript level of the target gene was significantly decreased and the lethal phe-notype were observed in N.lugens after feeding with dsRNA.We constructed the RNAi vector of the Nlst4 gene,which was then introduced into rice by Agrobacterium-mediated transformation method.After feeding with the transgenic rice,the transcript level of the target gene in N.lugens were significantly reduced,the 1st and 2nd instar nymph duration were significantly prolonged,but no lethal phenotype was observed.These re-sults indicated the practicability of breeding transgenic rice to control N.lugens by RNAi technique,and pro-vided the scientific basis for breeding transgenic rice for field-level control of planthoppers.关键词
