摘要

Abstract

Objective To observe the therapeutic effect of berberine on the treatment of radiation-induced intes-tinal injury and intestinal microbiota homeostasis via animal experiments and a small intestinal organoid model.Methods 80 male C57BL/6J mice aged seven weeks were randomly divided into four groups of the blank control group,radiation model group,amifostine group,and berberine group,with 20 mice in each group.In addition to the blank control group,the radiation-induced intestinal injury model was built by exposing the mice to a single dose of 8.5 Gy whole-body irradia-tion using 60 Co-γ-radiation.The amifostine group received intraperitoneal injection of amifostine half an hour before ra-diation,while the berberine group received daily doses of berberine via intragastric administration after radiation.Mean-while,this paper observed the survival status and recorded the average body weight of the mice.On the third day after radi-ation,staining was performed on small intestine sections to observe the status of the mucosa in small intestines.Additional-ly,fecal samples were collected from the mice for sequencing to analyze the effect of radiation and berberine on intestinal microbiota homeostasis.In the in vitro experiment,a cell model of radiation-induced intestinal injury was built by expo-sing small intestinal organoids to a similar dose of irradiation,which was added to the fecal supernatant in the berberine group for intervention.Meanwhile,this group was compared with the radiation model group and blank control group to ob-serve the effect of fecal supernatant in the berberine group on the growth status of small intestinal organoids after radiation.Results Compared with the radiation model group,the survival rate and average body weight of mice in the berberine group increased,with remarkable intestinal mucosa recovery.Meanwhile,compared to the amifostine group,the expression of Ki67 and Claudin in the small intestines notably increased(P<0.01),the abundance and a diversity of the intestinal flora in the berberine group were significantly increased(P<0.05),and the abundance of the beneficial bacteria was en-hanced.Specifically,the three beneficial bacteria of Bacteroides thetaiotaomicron,Bacteroides vulgatus,and Bacteroides acidifaciens which belong to the genus Bacteroides had significantly more abundance and accounted for a larger proportion in the total intestinal flora of the berberine group.In vitro experiments demonstrated that the fecal supernatant in the ber-berine group can significantly promote the growth of small intestinal organoids after radiation(P<0.01)and stimulate the expression of Lgr5 as a marker for stem cells in small intestines and Ki67 as a marker for cell proliferation(P<0.01).Conclusion Berberine can make intestinal microbiota homeostasis recover by increasing the abundance of Bacteroides,and create a micro-environment conducive to the proliferation of small intestinal mucosal epithelial cells,thus promoting the recovery of radiation-induced intestinal injury.
