Abstract

Objective To explore the metabolomic changes in the serum of dengue patients through comparative analysis of the metabolome of patients infected with dengue virus(DENV)and healthy people.Differential analysis and pathway analysis were performed to understand the host's metabolic response to the virus and to search for possible biomarkers.Method 19 dengue fever patients who visited Shenzhen Third Peoples Hospital from June 2023 to October were used as the research subjects,and 11 healthy people were used as controls.Their serum was collected for non-targeted metabolomic analysis.Use bioinformatics methods to conduct differential analysis and pathway analysis to screen differentially expressed metabolites and find host metabolic pathways affected by viral infection.Result Overall principal component analysis(PCA)and relative standard deviation(RSD)showed that the quantitative results of biological replicate samples were statistically consistent.Compared with the normal population,the serum metabolites of dengue patients in the experimental group were significantly different.A total of 114 metabolites showed significant expression differences(P<0.05,Fold Change ≥1.5 or ≤2/3).The HMDB annotation classification of differential metabolites showed that the differential metabolites were mainly concentrated on lipid metabolites.Conclusion The serum metabolomic profile of dengue patients significantly differs from that of healthy individuals,primarily characterized by alterations in lipid metabolism.These findings suggest that dengue virus infection modifies the host's lipid metabolism pathway,providing a theoretical foundation for understanding the pathogenesis of dengue virus infection and contributing to the diagnosis and prognosis of dengue fever.
