一株基因Ⅶ型新城疫病毒L蛋白致弱突变株的拯救及其免疫效力评价
Rescue and evaluation of immune efficacy of an attenuated strain of genotype Ⅶ Newcastle disease virus with L protein mutation
摘要
Abstract
In order to prevent and control the epidemic genotype Ⅶ Newcastle disease(ND),a genotypeⅦ Newcastle disease virus(NDV)was used as the skeleton,the amino acid sequence of its F protein cleavage site was mutated to the corresponding sequence of the La Sota strain and the lysine at position 1159 of its L protein to arginine,and an attenuated NDV mutant strain named as rNDV-Fmut-LK1159R with L protein mutation was rescued.The mutated sites of the rNDV-Fmut-LK1159R mutant strain can be genetically stable after continuous passage in chicken embryos up to the 15th generation.Four-week-old SPF chickens were immunized with the rNDV-Fmut-LK1159R mutant strain and the La Sota strain with 1 × 107 EID50 per bird once,respectively.And the chickens were challenged with a genotype Ⅶ NDV strain with 1 × 105 EID50 per bird on 21 days post immunization.The results showed that the rNDV-Fmut-LK1159R mutant strain could significantly increase the expression of IFN-γ mRNA in peripheral blood lymphocytes of SPF chickens,significantly stimulate the body to produce humoral and cellular immune responses,and provide complete immune protection for SPF chickens,effectively preventing virus shedding and inhibiting replication of the virus in their organs.In conclusion,the rNDV-Fmut-LK1159R mutant strain has excellent immunogenicity and has the potential to be an attenuated vaccine candidate of ND.关键词
