表达EGFP的重组LSDV的构建以及在高效筛选抗病毒药物中的应用
Construction of recombinant LSDV expressing EGFP and its application in high-efficient screening of antiviral drugs

Abstract
To effectuate high-content screening(HCS)of drugs against the lumpy skin disease virus(LSDV),the non-essential gene TK for LSDV replication was chosen as the EGFP insertion site to fabricate a recombinant LSDV expressing EGFP,denominated rLSDV-ΔTK-EGFP.The growth attributes of rLSDV-ΔTK-EGFP manifested no substantive disparity when compared with the wild-type LSDV.Grounded on rLSDV-ΔTK-EGFP,a swift high-content screening approach for drugs against LSDV was instituted.A cumulative total of 1 036 compounds from a natural product repository were screened employing this methodology.The outcomes demonstrated that 5 compounds possessing anti-LSDV activity were discerned.Further verification of LSDV117 protein expression levels and LSDV043 gene levels disclosed that the LSDV was conspicuously inhibited upon treatment with these 5 compounds on MDBK cells.The 5 compounds evinced no notable toxicity to MDBK cells at a concentration of 10 μmol/L.These findings suggest that the constructed recombinant LSDV expressing EGFP is applicable for high-content screening of anti-LSDV drugs and possesses a com-parably high screening efficiency.It holds considerable significance for enhancing the antiviral capability of individuals,mitigating the symptoms of lumpy skin disease,and minimizing economic losses.关键词
