Molecular Typing,Antimicrobial Resistance Characteristics,and Virulence Gene Detection of Staphylococcus aureus from Subclinical Mastitis in a Dairy Farm of Bayingol Mongolian Autonomous Prefecture,Xinjiang

Abstract
To investigate the molecular epidemiological characteristics of Staphylococcus aureus isolated from subclinical mastitis in dairy cows in Bazhou,Xinjiang Uyghur Autonomous Region,this study collected 40 milk samples from cows with subclinical mastitis in May 2020.Bacterial isolation and identification were performed,followed by multi-locus sequence typing(MLST)and staphylococcal protein A(spa)typing.Additionally,drug susceptibility,virulence gene profiles,and biofilm-forming abilities were analyzed.The results showed that 34 S.aureus strains were isolated from 40 milk samples.MLST analysis identified 18 sequence types,while spa typing revealed 7 types,with ST5405-t189 and ST5406-t705 being the predominant clones.Among the isolates,erythromycin resistance was the most prevalent(50.0%,17/34),and 4 methicillin-resistant S.aureus(MRSA)strains(11.8%)were detected.Six strains(17.6%)exhibited multidrug-resistant phenotypes.A total of 14 virulence genes were identified among the isolates,with high detection rates for clfB(94.1%,32/34),clfA(88.2%,30/34),hlb(85.3%,29/34),cna(79.4%,27/34),and hla(67.6%,23/34).Furthermore,23 isolates(67.6%)demonstrated biofilm-forming abilities.These findings indicate that the S.aureus isolates exhibit diverse molecular typing profiles,with some strains showing multidrug resistance,biofilm-forming capacities,and multiple virulence genes.The results highlight the need to enhance detection and control measures for S.aureus associated with subclinical mastitis in dairy cows.关键词
