表皮葡萄球菌多糖与牛支原体P48蛋白偶联物的免疫效力初步研究
Preliminary Study on the Immunogenicity of Staphylococcus epidermidis Polysaccharide-Mycoplasma bovis P48 Protein Conjugate

Abstract
This study aimed to evaluate the safety and immunogenicity of the polysaccharide(PS)of Staphylococcus epidermidis conjugated with the P48 protein of Mycoplasma bovis(PS-P48)in mice.Twenty BALB/c mice were randomly divided into four groups:Control,Polysaccharide,Protein,and Conjugate groups.Mice were immunized with 0.9%saline,PS,P48 protein,or PS-P48 conjugate,respectively,at 14-day intervals for three rounds of immunization.Clinical observations and body weight measurements were conducted on days 0,7,14,21,28,35,42,49,and 56 post-immunization.Serum samples were collected for antibody titration and cytokine analysis.The results showed no mortality or adverse reactions throughout the immunization period,with all groups maintaining good physical conditions.At 35,42,49,and 56 days post-immunization,the body weight of the Polysaccharide group was significantly lower than that of the Control group(P<0.05 or P<0.01).At 42 days,the body weight of the Protein group was significantly lower than that of the Control group(P<0.05).At 42,49,and 56 days,the body weight of the Conjugate group was significantly lower than that of the Control group(P<0.05 or P<0.01).In terms of antibody levels,the mean serum IgG P/N values of the Conjugate,Protein,and Polysaccharide groups were significantly higher than those of the Control group(P<0.01)at all time points post-immunization.The mean P/N values of the Conjugate and Protein groups were significantly higher than those of the Polysaccharide group(P<0.01).The mean P/N values of the Protein and Conjugate groups peaked at 49 days post-immunization.The Conjugate group's mean P/N value against the P48 protein was significantly higher than that of the Protein group(P<0.01),with antibody titers reaching 1∶25 600 in the Protein group and exceeding 1∶51 200 in the Conjugate group.Additionally,the antibody titer against PS in the Conjugate group was 1∶800,while the titer against P48 exceeded 1∶6 400.At 56 days post-immunization,serum IL-4,IL-6,and IFN-γ levels in the Conjugate group were significantly higher than those in the Protein,Polysaccharide,and Control groups(P<0.05 or P<0.01).These findings indicate that the PS-P48 conjugate can induce specific antibodies against both Staphylococcus epidermidis PS and Mycoplasma bovis P48 protein while activating cytokine expression.This provides valuable insights for further development of vaccines targeting both Staphylococcus epidermidis and Mycoplasma bovis,particularly for the prevention of bovine mastitis.关键词
