基于AMPK/MTOR/ULK1信号通路探讨头针治疗脑出血的作用机制
Mechanism of Scalp Acupuncture in Treating ICH Based on AMPK/MTOR/ULK1 Signaling Pathway

Abstract
Objective:To explore the mechanisms of scalp needling Baihui(DU20)penetrating Qubin(GB7)in treatment of intracerebral hemorrhage(ICH)in rats based on AMPK/MTOR/ULK1 signaling pathway.Methods:The 81 male SD rats of SPF grade were randomly divided into the sham group,the model group and the acupuncture group,and each group was subsequently divided into 3 subgroups of 1 d,3 d and 7 d,with 9 rats in each subgroup.The ICH model was established by autologous blood injection method.The acupuncture group was treated with scalp needling DU20 penetrating GB7,once every 24 h.The rats in the sham group and the model group were bound and immobilized without treatment during the treatment period of the acupuncture group.Ludmila Belayev score was used to evaluate the neurological function of rats in each group;the morphological changes of brain tissue were observed by HE staining;the expressions of GPX4,MDA and GSH in the brain tissues were measured by ELISA;the expressions of AMPK,P-AMPK,MTOR,P-MTOR,ULK1,P-ULK1,LC3B and Beclin1 in the brain tissues were detected by WB.Results:The rats in the model group showed neurological deficits at all times,and the difference was statistically significant compared with that in the sham group(P<0.01).The neurological deficits in the model group were found to be most severe at the 3rd day of ICH when compared to other time points.The neurological function was improved at all time points in the acupuncture group compared with that in the model group after 3 and 7 days of consecutive treatment(P<0.05).The arrangement of brain tissue cells was significantly looser and irregular,the neurons were significantly reduced and the cells were swollen in the model group compared with those in the sham group at the same time.The structural damage of brain tissues was the most obvious on the 3rd day after ICH and a large number of inflammatory cells were infiltrated,which were alleviated on the 7th day after ICH in the model group.The degree of damage to the surrounding tissues of ICH location was significantly improved after acupuncture intervention.The expressions of GPX4 and GSH in brain tissues were decreased(P<0.01),and MDA was increased after ICH(P<0.01);acupuncture was seen to improve their expressions in the brain at different time points(P<0.01).After ICH,phosphorylation activation of AMPK,MTOR and ULK1 occurred in brain tissues,in which P-AMPK,P-ULK1,LC3B and Beclin1 were significantly decreased(P<0.05),whereas P-MTOR was increased(P<0.05),and the acupuncture treatment improved the expression results of the above factors.Conclusion:Scalp acupuncture attenuates ferroptosis in tissues surrounding lesions of ICH and promotes AMPK/MTOR/ULK signaling pathway-mediated cellular autophagy in brain tissues to protect neurological function of ICH rats.关键词
