天山雪莲提取物的保肝活性研究
Research on Hepatoprotective Activity of Saussureae Involucratae Herba Extract

Abstract
Objective:To explore the protective effect of Saussureae Involucratae Herba extract on the liver.Methods:The hepatocyte injury model induced by acetaminophen(APAP),oxidative hepatocyte injury model induced by hydrogen peroxide(H2O2),lipid deposition model in human hepatocytes induced by sodium oleate(NaOL),and activation model of hepatic stellate cells of mice induced by transforming growth factor-β(TGF-β)were established.The cell survival rate and inhibition rate were detected by MTT assay,so as to evaluate the activity of Saussureae Involucratae Herba extract on hepatocyte injury,hepatic lipid deposition,and hepatic fibrosis in vitro.The model of mice with immune liver injury induced by concanavalin A(Con A)and the model of oxidative liver injury induced by carbon tetrachloride(CCl4)were prepared.The therapeutic effect of Saussureae Involucratae Herba extract on liver injury in vivo was evaluated by measuring organ index,the contents of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in serum,and malondialdehyde(MDA)and reduced glutathione(GSH)in liver tissue.Results:Saussureae Involucratae Herba extract showed remarkable hepatoprotective activity in vitro,significantly increasing the survival rate of cells,reducing lipid deposition in hepatocytes,and inhibiting the activation of hepatic stellate cells induced by TGF-β.In vivo studies revealed that Saussureae Involucratae Herba extract showed good hepatoprotective activity in mice with immune liver injury and those with oxidative liver injury.Saussureae Involucratae Herba extract of 1 000 mg·kg-1 could reduce AST level in serum(P<0.05)and MDA level in liver tissue(P<0.05)in mice with acute immune liver injury;Saussureae Involucratae Herba extract of 500 mg·kg-1 could significantly reduce ALT level in serum(P<0.05),lower MDA level in liver tissue(P<0.05),and increase GSH level in liver tissue(P<0.05)in mice with oxidative liver injury.Conclusion:Saussureae Involucratae Herba extract shows protective activity on liver injury caused by different causes,and it can reduce lipid deposition in hepatocytes and inhibit the activation of hepatic stellate cells.It has certain potential in the treatment of liver diseases,and anti-oxidation may be one of its mechanisms.关键词
