Abstract

Objective To study the optimal extraction process of total saponins from Mongolian Astragalus in Mongolia.Methods The ethanol reflux extraction was used to determine the optimal extraction process of Mongolian Astragalus using the total saponin yield of Mongolian Astragalus as an index,and the effects of four factors,namely,ethanol concentration,ethanol dosage,extraction time,and num-ber of times of extraction,were investigated by a one-way combination of Box-Behnken response surface method,and the regression model was established and validated to determine the optimal extraction process of Mongolian Astragalus.Results The regression model was significant and the fitting accuracy was high;the order of influencing the reflux extraction of total saponins from Mongolian Astragalus was ethanol dosage>ethanol concentration>extraction times>extraction time,ethanol concentration and ethanol dosage were signifi-cant,and more total saponins from Mongolian Astragalus were obtained with 70%ethanol concentration and 25 times of ethanol dosage,while the number of extractions and the time of extraction were not significant.Conclusion The optimum extraction process conditions for Mongolian Astragalus were 70%ethanol concentration,3 times extraction times,25 times ethanol dosage and 90 min extraction time.The yield of total saponins of Mongolian Astragalus extracted under this process was 3.201%.The response estimates were in good agree-ment with the actual valuse.
