Abstract
Objective:Pancreatic ductal adenocarcinoma(PDAC)is highly malignant with limited opportunities for curative surgery,and traditional chemotherapy is not very effective.This study aims to explore the expression of Claudin18.2 protein in PDAC and its correlation with clinical pathological features,in an attempt to provide new insights for targeted therapy of PDAC.
Methods:The expression of Claudin18.2 was detected using the Optiview DAB detection kit on the Ventana platform in 30 normal pancreatic tissues and 310 PADC tumor tissues,along with corresponding metastatic lymph nodes.The relationship between the expression level of Claudin18.2 and the clinical pathological features of PDAC was analyzed,along with the consistency of Claudin18.2 expression between primary PDAC lesions and metastatic lymph nodes.
Results:Claudin18.2 was negative in all normal pancreatic tissues(0/30)and showed a positive rate of 74.5%(231/310)in PDAC,with 48.1%(149/310)showing high expression.The expression level of Claudin18.2 was significantly associated with the presence of lymph node metastasis and nerve invasion(both P<0.05).High expression of Claudin18.2 was significantly increased in PDAC with lymph node metastasis and nerve invasion.However,the expression level of Claudin18.2 was not related to patient gender,age,tumor location,maximum tumor diameter,or differentiation degree(all P>0.05).In the lymph node metastases of PDAC,the positivity rate of Claudin18.2 was 68.6%(129/188),with a high expression rate of 48.9%(92/188).The consistency of Claudin18.2 expression levels between the primary PDAC lesions and lymph node metastases was strong(Kappa value=0.819,P<0.05).
Conclusion:Claudin18.2 has a high positive rate and high expression rate in PDAC.The expression consistency between primary lesions and metastatic lymph nodes is high,suggesting that Claudin18.2 may be a potential target for zolbetuximab treatment in PDAC.关键词
