MSI1在乳腺恶性肿瘤中作用机制的研究进展
Research progress on the mechanism of MSI1 in breast malignant tumors

Abstract
Musashi1(MSI1)is an RNA-binding protein that regulates post-transcriptional target gene expression by binding to specific recognition sequences,activating or inhibiting the translation of target mRNAs.As a stem cell marker,MSI1 is highly enriched in normal and tumor stem cells in organs or systems such as the intestine,central nervous system,and reproductive system,maintaining stem cell characteristics,promoting tumor metastasis,and being associated with poor prognosis.The expression of MSI1 in breast tumors is higher than in normal breast tissue,and it is associated with metastasis and prognosis.MSI1 maintains breast cancer stem cell characteristics by negatively regulating p21.MSI1 binds to the 3'-untranslated region(3'-UTR)of Numb mRNA to prevent the translation of the Notch inhibitor Numb;it binds to the mRNA of the transcription factor NF-YA to mediate the downregulation of the 26s proteasome,preventing the degradation of the intracellular domain of Notch and maintaining Notch signaling,thereby regulating the self-renewal of breast cancer stem cells.Moreover,MSI1 is involved in the progression,metastasis,epithelial-mesenchymal transition inhibition,extracellular matrix degradation,and chemotherapy resistance in breast cancer,showing potential as a target for radio-sensitization.Based on the role of MSI1 in the occurrence and development of breast malignancies,MSI1 is expected to become a new therapeutic target for breast cancer.关键词
