The role of histone deacetylase 6 in diabetes mellitus and its complications

Abstract
Diabetes mellitus(DM)is a metabolic disorder characterized by persistent hyperglycemia,leading to damage of various tissues and organs such as muscles,brain,eyes,kidneys,heart,and blood vessels,thereby causing multiple complications including diabetic nephropathy(decline in glomerular filtration function),acute diabetic heart injury(myocardial infarction),diabetic cognitive dysfunction(memory loss,inattention,slow thinking,and reduced learning ability),diabetic muscle atrophy(muscle tissue atrophy and weakened muscle strength),and diabetic eye disease(retinopathy,decreased vision,and ocular symptoms).Studies have implicated the dysregulation of histone deacetylase(HDAC)in various human diseases,including DM.Histone deacetylase 6(HDAC6)is a distinctive member of the histone deacetylase family,characterized by its cytoplasmic localization and selective deacetylation of cytoplasmic proteins,distinguishing it from other family members.Numerous studies indicate an upregulation of HDAC6 expression and activity in neurodegenerative diseases,acute liver injury,and animal models of tumors.Moreover,research indicates the significant involvement of HDAC6 in DM and its complications,with HDAC6 inhibitors effectively delaying the advancement of diverse metabolic diseases,indicating that targeting HDAC6 expression or activity could offer a novel therapeutic avenue for DM treatment.This article reviews the distinctive functions of HDAC6 and outlines the potential of its inhibitors in treating DM and its complications.关键词
