基于分子标记技术的马尾松和火炬松杂交子代鉴定与抗性评价
Identification and Resistance Evaluation of Hybrid Offspring of Pinus massoniana and Pinus taeda Based on molecular Marker technology

Abstract
[Purpose]The early,rapid and accurate identification of hybrid offspring between Pinus massoni-ana and P.taeda,and the breeding of high resistant P.massoniana×P.taeda family are of great signific-ance for the prevention and control of pine wilt disease.[Method]This study used P.massoniana as the female parent and P.taeda as the male parent for hybridization.Specific primers were designed by com-paring the genome sequences of P.taeda and P.tabulaeformis.Specific primers between P.massoniana and P.taeda were screened,and amplification band analysis was performed on the hybrid offspring to es-tablish a rapid identification system for P.massoniana×P.taeda.Families with high resistance were eval-uated and screened by inoculating with pine wood nematodes.[Result]12 primer combinations were se-lected from 150 pairs for identification,with strong specificity,good stability,and high amplification rate.A total of 88 samples were collected from 29 families in the F1 generation,of which 81 were identified as hy-brid plants with a hybrid rate of 92.05%;There were a total of 688 samples from 21 families in the F2 gen-eration,of which 279 were hybrid plants with a hybrid rate of 40.55%;5 highly resistant families were iden-tified from the resistance evaluation of 13 families in the F2 generation of P.massoniana×P.taeda,result-ing in.[Conclusion]The 12 selected primer combinations effectively authenticate hybrid offspring of P.massoniana and P.taeda.Furthermore,5 highly resistant families were identified,providing a foundation for pine wilt disease prevention,control,and further research into resistance mechanisms.关键词
