摘要
Abstract
Objective:To investigate the effects of curcumin(CUR)on the activity and angiogenesis of dental pulp stem cells by regulating the hypoxia inducible factor-1α(HIF-1α)/vascular endothelial growth factor(VEGF)signaling path-way.Methods:Human dental pulp stem cells(hDPSCs)were cultured in vitro and randomly separated into Control group(normal culture),L-CUR,M-CUR,H-CUR groups(125,250,500 nmol/L CUR),and CH group(500 nmol/L CUR+100 µg/mL HIF-1α/VEGF signaling pathway inhibitor Chloramphenicol).MTT method and plate cloning method were applied to detect cell proliferation in each group.Scratch experiment was applied to detect the migration ability of cells in each group.The vas-cular mimicry formation experiment was applied to detect the potential of tube formation.Western blotting(WB)was used to detect the expression of proliferating cell nuclear antigen(PCNA)and HIF-1α/VEGF signaling pathway related proteins(HIF-1α,VEGF)of cells in each group.Results:Compared with the Control group,the A490 of hDPSCs cells,plate clone count,scratch healing rate,tube forming potential,PCNA,HIF-1α,and VEGF protein expression in the L-CUR group,M-CUR group,and H-CUR group increased(P<0.05).Compared with the H-CUR group,the A490,plate clone count,scratch healing rate,tube forming potential,PCNA,HIF-1α,and VEGF protein expression in the CH group decreased(P<0.05).Conclu-sion:CUR promotes angiogenesis in hDPSCs cells by activating the HIF-1α/VEGF signaling pathway,thereby promoting cell proliferation and migration.关键词
