Abstract
Objective:To explore the effects of serum from patients with primary Sjögren's syndrome(pSS)on ferrop-tosis and inflammation of A253 cells.Methods;Transmission electron microscopy was carried out on the labial gland tissues of patients with primary Sjögren's syndrome and normal volunteers to observe the ultrastructure of the samples.Perform Per-form HE staining on their tissue specimens to observe the content of collagen fibers in the tissues,and conduct Perl's staining to observe the content of hemosiderin in the tissues.The primary Sjögren's syndrome and normal volunteers were used to stim-ulate A253 cells.After 24 hours of stimulation,RT-qPCR and Western blot were used to detect the level of transcription and protein of ferroptosis-related indicators such as long-chain acyl-CoA synthetase 4(ACSL4),glutathione peroxidase 4(GPX4),ferritin heavy chain 1(FTH1)and so on,as well as related inflammatory factors such as interleukin(IL)-1 β,IL-6,IL-17,etc.The detection of reactive oxygen species and Liperfluo was used to assess the lipid peroxidation level of the cells after serum stimulation.Subsequently,the ferroptosis inhibitor Ferrostatin-1(2 µmol/mL)was added to the pSS serum-trea-ted,and the changes in the above ferroptosis and inflammation-related indicators were observed.Results;Obvious ferroptosis-related indications were observed in the labial gland tissues of patients with Sjögren's syndrome and in the groups treated with the serum of patients with Sjögren's syndrome.Moreover,compared with the groups treated with the serum of normal volun-teers,the levels of ferroptosis,lipid peroxidation level,and the expression of inflammatory factors were all increased in the groups treated with the serum of patients with Sjögren's syndrome.Also,the addition of Ferrostatin-1 could reduce the impact of the serum of patients with Sjögren's syndrome on the above indicators.Conclusion:Stimulation by pSS serum will exacer-bate ferroptosis and the production of inflammatory factors in A253 cells.关键词
