Based on bioinformatics and experimental verification,a diagnostic model established of circadian rhythm and periodontitis related genes

Abstract
Objective:To investigate the role of circadian rhythm related genes in the occurrence and development of periodontitis through bioinformatics analysis and experimental verification of circadian rhythm hub genes associated with peri-odontitis.Methods:Use R software to screen differentially expressed circadian rhythm related genes in periodontitis and per-form enrichment analysis to identify key pathways and genes.Conduct qRT-PCR experiments on saliva samples collected from periodontitis and healthy patients to verify the expression of hub genes.Perform COX regression analysis on hub genes and construct a diagnostic model for periodontitis related to circadian rhythm to explore its diagnostic value for periodontitis.Re-sults:Eight hub genes related to the circadian rhythm of periodontitis were screened,and qRT-PCR results of saliva samples showed that the relative expression of the eight hub genes was higher in periodontitis patients than in the control group,with statistical significance consistent with bioinformatics results.GO analysis found that these genes are mainly involved in the positive regulation of lipopolysaccharide mediated signaling pathways,the regulation of intrinsic apoptosis signaling pathways induced by endoplasmic reticulum stress,and the differentiation of mature B cells in immune responses.A diagnostic model of circadian rhythm-related periodontitis was constructed based on stepwise regression and COX regression analysis of eight hub genes,and the AUC value showed that the model had good diagnostic performance and had potential application value in the clinical diagnosis of circadian rhythm-related periodontitis.Conclusion:This study has identified a potential link between pe-riodontitis and circadian rhythm at the genetic level,which helps to reveal the mechanism of action between circadian rhythm disorder and periodontitis,and thus explore new temporal medical treatment methods for the prevention and treatment of peri-odontitis.关键词
