基于多指标评价巴戟天药材质量
Medicinal material quality evaluation of Bajitian based on multiple indexes

Abstract
Objective:To determine the contents of 7 compounds in 16 batches of Bajitian samples by high-performance liquid chromatography-ultraviolet detection(HPLC-UV),evaluate the quality of Bajitian by grey correlation degree analysis,membership function method and chemical pattern recognition method,and perform quality difference analysis.Methods:The contents of 4 iridoid glycosides and 3 anthraquinones in 16 batches of Bajitian samples were deter-mined by HPLC-UV.The quality of Bajitian was comprehensively evaluated by grey correlation degree analysis and mem-bership function method,and its quality difference was analyzed by chemical pattern recognition method.Results:The linear ranges of monotropein,deacetyl heliosylic acid,chlorogenic acid,asperuloside,2-hydroxy-1-methoxy anthraquinone,rubia-din-1-methyl ether and rubiadin were 0.2775~2.7750 μg,0.1050~1.0500 μg,0.0060~0.0600 μg,0.0017~0.0170 μg,0.0048~0.0960 μg,0.0096~0.1920 μg and 0.0100~0.2000 μg,respectively.The linear relationship was good.The contents of the above 7 components in 16 batches of Bajitian samples were 4.363~22.213 mg/g,2.479~7.527 mg/g,0.068~0.276 mg/g,17.690~161.757 μg/g,0.966~108.064 μg/g,1.316~135.488 μg/g,and 1.639~29.044 μg/g,respectively.The equal-weight relative correlation degree of 16 batches of Bajitian samples ranged from 0.28 to 0.60.The weighted relative correlation ranged from 0.15 to 0.71.The total average membership values ranged from 0.07 to 0.68.These results indicated that there were some quality differences among different batches of Bajitian samples.The 16 batches of Bajitian samples were clus-tered into two classes by cluster analysis.S1 alone was clustered into one class,and the other samples were clustered into one class.Three principal components were extracted by principal component analysis,and the cumulative variance contri-bution rate was 96.9%.And rubiadin-1-methyl ether,rubiadin,and 2-hydroxy-1-methoxy anthraquinone were screened as main chemical markers for the quality difference of Bajitian by Orthogonal Partial Least-Squares Discrimination Analysis.Conclusion:There are some differences of iridoid glycosides and anthraquinones in Bajitian samples from different areas with different grown years,which may be related to the growing environment,growing years and germplasm resources.关键词
