Target gene prediction and bioinformatics analysis of bta-miR-134

Abstract
In this study,the RNA samples of mammary epithelial cells of cows co-treated with LPS and calcium ions as well as the RNA samples of mammary epithelial cells of cows treated with LPS were subjected to miRNA sequencing.Sixteen differentially ex-pressed miRNAs were identified,of which one exhibited increased expression level and fifteen displayed decreased expression.The miRNA that up-regulated was bta-miR-134,which may play a pivotal role in the inhibition of the inflammatory response process of cow mammary epithelial cells by calcium ions.Accordingly,the target genes of bta-miR-134 derived from cow mammary epithelial cells were predicted,and bioinformatics analysis was conducted to establish a research foundation for the functional verification of bta-miR-134.The miRBase database was employed for the analysis of the maturation sequence conservation of bta-miR-134.In or-der to predict the target genes of bta-miR-134,the Targetscan database was utilised.The clusterProfiler package in R was employed to conduct GO and KEGG enrichment analyses of the target genes of bta-miR-134.The STRING database was employed for the analysis of protein interactions among the target genes of bta-miR-134.The mature sequence of bta-miR-134 exhibits high conserva-tion among different species.A total of 171 target genes of bta-miR-134 were predicted.The enrichment results of GO demonstrated that the target genes were predominantly enriched in protein serine/threonine kinase activity,protein kinase activity,protein serine kinase activity and other molecular functions.The KEGG pathway enrichment results indicated that the target genes were predomi-nantly associated with the Ras signaling pathway,neurotrophic factor signaling pathway,human immunodeficiency virus type 1 in-fection and Chagas disease.The protein interaction analysis revealed that KRAS,PIK3CA and CASP8 exhibited the highest degree of connectivity with other genes.Bta-miR-134 may target key genes,such as KRAS,PIK3CA and CASP8,and is further mediated by the Ras signaling pathway,the MAPK signaling pathway and the Toll-like receptor signaling pathway,thereby regulating the in-flammatory response of cow mammary epithelial cells.This study provides an important data base for further revealing the regulato-ry mechanisms between calcium ion,miRNA and inflammatory signaling pathways in mammary epithelial cells of cows.关键词
