Regulatory Effects of Calcium and Non-Myosin on Apical Opening of Zebrafish Epidermal lonocytes

Abstract
In order to study the factors affecting the size of the apical opening of ionocytes,this study used zebrafish as a model organism to explore the factors by immunofluorescence and drug treatment.The apical opening was usually investigated via electron microscopy,which is relatively laborious.In this study,we reported a simple method to visualize apical opening of ionocytes via antibody staining of Claudin-5,a tight junction protein.We analyzed the apical opening of ionocytes using several small mol-ecule compounds.Our data showed that treatment of zebrafish embryos with calcium inhibitors reduced the size of apical opening in ionocytes.Similarly,incubation with inhibitor of non-muscle myosin Ⅱ also resulted in the decreased size of apical opening.In contrast,treatment with either epinephrine or thyroid hormones had no effect on the apical opening of ionocytes.These results suggested that the apical open-ing of ionocytes is controlled by a calcium dependent regulatory mechanisms which may require the in-volvement of non-muscle myosin Ⅱ.Our data may provide further information for the study of the ho-meostasis and physiological function of ionocytes.关键词
