Abstract

The experiment aimed to explore the optimal extraction process of Ligusticum acuminatum Franch(LAF)polysaccharides as well as their antioxidant and antibacterial activities.The extraction of LAF polysaccharides was conducted using an ionic liquid-assisted microwave extraction method.The optimal extraction process was determined through single-factor experiments and response surface optimization.After steps including defatting,decolorization,protein removal,dialysis,and purification,a uniform acidic polysaccharide(LAF-3)was obtained.The antioxidant and antibacterial effects were evaluated through in vitro tests.The results showed that when the microwave power is 300 W,the microwave time was 60 min,the liquid material ratio was 40 mL/g,the microwave temperature was 60 ℃,and the mass concentration of ionic liquid was 15 g/L,the highest extraction rate of LAF-3 was 41.481%.The IC50 of LAF-3 for scavenging 1,l-diphenyl-2-trinitrophenylhydrazine(DPPH)radical and total reducing capacity were 7.491 and 8.261 g/L.LAF-3 had a good inhibitory effect on Escherichia coli,Staphylococcus aureus and Candida albicans,the diameters of the inhibition zones were 12.13,11.35,and 10.08 mm respectively.The study indicates that the LAF-3 had good in vitro antioxidant and antibacterial ability.
