Abstract

The aim of this experiment was to optimize the freeze-drying process of Lactiplantibacillus plantarum HYY-DB9.Using Lactiplantibacillus plantarum HYY-DB9 as the research subject,the study selected optimal centrifugation conditions,types and concentrations of freeze-drying protectants,and storage temperature and time.Through orthogonal experiments,a composite protectant formula was determined,and the storage stability of HYY-DB9 was verified.The results showed that when the centrifugation condition was set at 8 000 r/min for 10 minutes,the viable cell count of Lactiplantibacillus plantarum HYY-DB9 reached the maximum,with a survival rate of(92.39±5.84)%.The optimal composite protective agent formula was as follows:The addition amount of bacteriological peptone was 4%,the addition amount of trehalose was 8%,and the addition amount of sodium glutamate was 6%.Under this condition,the survival rate of Lactiplantibacillus plantarum HYY-DB9 reached(94.67±1.89)%,and the survival rate of Lactiplantibacillus plantarum HYY-DB9 stored at-20 ℃ for 28 days was(86.94±0.74)%.The study indicated that this protective agent formula had a good protective effect on the freeze-drying and storage process of Lactiplantibacillus plantarum HYY-DB9.
