Comparison of transcriptome sequencing between Nubian goats and Hu sheep under heat stress

Abstract

The experiment aimed to investigate the differences and similarities in differentially expressed genes(DEGs)and pathways related to muscle growth and development between Nubian goats and Hu sheep under heat stress(HS)conditions.Transcriptome sequencing(RNA-Seq)was used to sequence the muscles of Nubian goats and Hu sheep,respectively,and to compare and analyze the differences in RNA-Seq results.The results showed that a total of 20 587 genes were detected in the goat group,with 95 DEGs identified,including 34 up-regulated genes and 61 down-regulated genes.In the Hu sheep group,a total of 14 685 genes were identified,with 180 DEGs selected,including 71 up-regulated genes and 109 down-regulated genes.GO functional enrichment analysis indicated that HS significantly affected 509 functional categories in the goat group,with DEGs mainly involved in binding,biological system regulation,and immune system-related functions.HS significantly affected 731 functional categories in the Hu sheep group,with DEGs mainly involved in binding,protein activity,and extracellular matrix-related functions.KEGG pathway enrichment analysis showed that the goat group was involved in 156 pathways,with DEGs mainly enriched in cell adhesion molecules,MAPK signaling pathways,phospholipase D signaling pathways,and focal adhesion signaling pathways,from which five DEGs potentially involved in muscle development were further selected:MET proto-oncogene receptor tyrosine kinase(MET),mitogen-activated protein kinase 10(MAPK10),fibroblast growth factor 1(FGFi),phospholipase A2 group IVE(PLA2G4E),and dual-specificity phosphatase 1(DUSP1);the Hu sheep group DEGs were involved in 164 pathways,with DEGs significantly enriched in calcium signaling pathways,ECM-receptor interaction,PPAR signaling pathways,and focal adhesion signaling pathways,and seven DEGs related to muscle development were selected:Chondroadherin(CHAD),P-selectin precursor(SELP),type Ⅰ collagen α1 chain(COL1A1),type Ⅰ collagen α2 chain(COL1A2),type Ⅵ collagen α6 chain(COL6A6),integrin-11(ITGA11),and prostaglandin F receptor(PTGFR).The study indicates that HS affects the main pathways and regulatory genes involved in muscle growth and development differently in Nubian goats and Hu sheep,but focal adhesion is a common pathway affected by HS in the muscle growth and development of both goats and sheep.
