Abstract
The experiment aimed to explore the fermentation conditions for the production of Monacolin K by the Monascus ruber BM0321 in liquid fermentation.The trial employed ambient temperature and pressure plasma mutagenesis technology to mutate the original strain,Monascus ruber BM0157,and selected a high-yielding and stable strain.Subsequently,single-factor experiments combined with response surface methodology were used to optimize the trial to determine the optimal fermentation conditions for Monascus ruber BM0321.The results showed that under the conditions of 145.42 g/L glycerol,261.6 mL/L soybean hydrolysate,80 g/L rice powder,2.1 g/L ZnSO4·7H2O,5 g/L corn steep powder,pH 5.5,10%inoculum size,and 20%loading volume,the Monacolin K content could reach 1 093.5 mg/L through shake flask fermentation.In a 100 L fermenter batch fermentation trial with Monascus ruber BM0321,the Monacolin K content could reach 848.23 mg/L under these conditions.The study indicates that the results can provide a reference for the industrial production of Monacolin K by liquid fermentation of Monascus ruber BM0321.关键词
