不同贮存方式对大豆种子活力及相关指标的影响

Abstract

In order to investigate the impact of different storage methods on the vigor and related indicators of soybean seeds under the high temperature and high humidity climate conditions of the south,two spring soybean cultivars'Guichun No.8'(GC 8)and'Guichun No.13'(GC 13),and one summer soybean cultivar'Guixia No.1'(GX 1)were used as experimental materials.Three treatments,sealed and dried condition(A1),sealed condition(A2),and natural storage(A3)were set up and placed under natural conditions(average daily temperature:8.2-29.9 ℃,average daily humidity:55.3%-88.0%)for 1 year with storage at-18 ℃(A0)as the control.The seed vigor,enzyme activities of peroxidase(POD),superoxide dismutase(SOD),and catalase(CAT),as well as malondialdehyde(MDA)content were measured.The electrical conductivity(EC)of the soaking solution and the weight of exuded solids(WDS)after soaking the seeds for 12 h were determined.The germination-related indicators of different storage treatment soybean seeds,and germination-related indicator of different storage treatment soybean seeds after drought treatment(DT)and flooding treatment(FT)were measured by using paper bed method.The results indicated that:1)In the TTC test,the color rendering index(CRI)of cotyledons for three cultivars in A0,A1,and A2 were all above 90%.The CRI of cotyledons for'GC 8'and'GC 13'seeds treated with A3 were reduced to 29.46%and 26.05%,respectively,while the CRI of cotyledons for'GX 1'seeds was reduced to 0.2)Compared to A0,A1,and A2,the electrical conductivity(EC)and weight of exuded solids(WDS)for A3 of the three cultivars were the highest.The WDS of the three cultivars in A1 was not significantly different from those in A0.Except for the EC of'GC 8'in A,was significantly higher than that in A0,there was no significant difference to EC between A1 and A0 for the other two cultivars.3)The MDA content of three cultivars in A2 was significantly higher than those in A0.The SOD content of three cultivars in A0 and A1 was significantly higher than that in A3,and the POD content of three cultivars in A0 and A,were significantly higher than those in A2 and A3.The CAT of three cultivars in A1,A2 and A3 were no significant difference from those in A0.4)The four indices of germination potential percentage(GPP),germination percentage(GP),normal seedling percentage(NSP),and germination index(Gl)of three cultivars in A3 were all reduced to 0.The four indices in A2 were significantly lower than those in A0 and A1.Compared to A0,in the four germination indicators of A1,except for the NSP of'GC 13'and the GPP and NSP of'GC 8'which were significantly lower than A0,there were no significant differences in the others.Compared with A0,the 5 d root length(5dRL),hypocotyl length(HL),root fresh weight(FRW),and vigor index(Ⅵ)for three cultivars in A2 were all lower.5dRL and Ⅵ of three cultivars in A2 were lower than those in A1.Except for the 5dRL of'GC 13'in A1,which was significantly lower than that in A0,the other cultivars in A1 showed no significant difference from those in A0.The root elongation rate(RER)of the three cultivars in A,and A2showed no significant difference compared to those in A0.5)Compared with A0,the GPP,GP,NSP,and Gl of the seeds in A1 and A2 after FT and DT stress treatment were reduced significantly,after FT and DT stress treatments,the extent of reduction in the four indicators of the three cultivars was different.In the FT,the reduction in GPP,GP,NSP,and Gl for'GC 13'and'GX 1'in A,and A2 was higher than that in A0.In the DT,the reduction of GP and NSP in A1 and A2 for all three cultivars was higher than that in A0.In summary,soybean seeds treated with different storage methods all exhibited varying degrees of aging after being stored under natural conditions for 1 year.Among these,sealed and dried conditions,(A1)showed the least degree of deterioration,followed by sealed storage(A2),with natural storage(A3)showing the highest degree of deterioration.Therefore,the sealed dry storage method can maintain the seed vigor of soybean in production under high temperature and high humidity condition in the storage in southern of China.
