Abstract

OBJECTIVE To investigate the effect of electroacupuncture on ocular dominance plasticity in adult amblyopic mice.METHODS The study was conducted in two experimental parts.In Experiment 1,10 eight-week-old adult mice underwent monocular deprivation(MD)of the right eye for four consecutive days and were then randomly assigned to the electroacupuncture(EA)group or the sham electroacupuncture(Sham)group(five mice per group).The EA group received electroacupuncture at Baihui(GV20),Yamen(GV15),and bilateral Hegu(LI4),while the Sham group underwent shallow needle insertion at the same acupoints without electrical stimulation.In Experiment 2,15 three-week-old mice underwent MD of the right eye until adulthood(eight weeks old),rendering the right eye amblyopic.These mice were randomly divided into the model group(MG),occlusion group(OG),and electroacupuncture+occlusion group(EAOG),with an additional blank control group(BG)(five mice per group).The BG and MG groups received no special interventions.In the OG group,the right eye was reopened at eight weeks of age,and the dominant left eye was occluded for seven consecutive days.The EAOG group underwent electroacupuncture intervention based on the OG protocol,following the same EA procedure as in Experiment 1,for seven consecutive days.Ocular dominance plasticity was assessed using in vivo electrophysiological recordings of the visual cortex.The contralateral(C)and ipsilateral(I)eye response values were used to calculate the C/I ratio for cell classification and to determine the contralateral bias index(CBI).In Experiment 1,the cumulative distribution of the ocular dominance index was compared between the two groups.RESULTS(1)Cell type distribution in Experiment 1:There was a significant difference in cell type distribution between the two groups(χ2=39.622,P=0.000).(2)CBI in Experiment 1:The CBI of the EA group was significantly lower than that of the Sham group(t=-4.889,P=0.001).(3)Cumulative distribution of ocular dominance index in Experiment 1:The EA group exhibited a significant shift in ocular dominance toward the ipsilateral eye compared to the Sham group(D=0.857,P=0.000).(4)Cell type distribution in Experiment 2:There was a significant difference in cell type distribution between the BG and MG groups(χ2=103.360,P=0.000).Significant differences were also observed between the MG group and both the OG(χ2=36.210,P=0.000)and EAOG(χ2=98.750,P=0.000)groups.Additionally,the OG and EAOG groups showed a significant difference in cell type distribution(χ2=41.800,P=0.000).(5)CBI in Experiment 2:The CBI of the MG group was significantly lower than that of the BG group(t=12.580,P=0.000).Both the OG and EAOG groups had significantly higher CBI values compared to the MG group(tOG=4.713,P=0.008;tEAOG=9.021,P=0.000),and the EAOG group exhibited a significantly higher CBI than the OG group(t=4.903,P=0.006).CONCLUSIONS Electroacupuncture can enhance visual cortex plasticity in adult amblyopic mice and promote the recovery of abnormal ocular dominance distribution.
