橙皮苷对模型小鼠肺纤维化的改善作用
Improvement Effect of Hesperidin on Pulmonary Fibrosis in Model Mice

Abstract
Objective To investigate the improvement effect and mechanism of hesperidin on the pulmonary fibrosis(PF)in model mice.Methods A total of 60 mice were randomly divided into the normal control group(gavage,equal volume of physiological saline),the model group(gavage,equal volume of physiological saline),the low-and high dose-hesperidin groups(gavage,25 mg/kg,100 mg/kg),the oe-EI24 group(one-time tail vein injection,16 µg,0.8 mL),and the oe-NC group(one-time tail vein injection,16 µg,0.8 mL),with 10 mice in each group.Mice were nasal fed with bleomycin(0.1 U/25 mg,20 µL)to replicate the PF model,while the mice in the normal control group were nasal fed with an equal volume of physiological saline.After 14 d of modeling,the mice in each group were given corresponding drugs by gavage once a day for 7 d.Hematoxylin-eosin staining and Masson staining were used to observe the morphology of mouse lung tissue under the microscope;the immunohistochemistry was used to detect the expression level of Collagen Ⅲ;the Western blot were used to detect the expression levels of corresponding factor proteins.Results Compared with those in the model group,the mice in the high-and low-dose hesperidin groups showed less lung collapse,improved lung tissue morphology,less collagen deposition and fibrosis formation;the inflammatory score and pulmonary fibrosis score significantly decreased(P<0.05).Compared with the those in model group,the expression levels of Collagen Ⅲ,Collagen I,Vimentin and α-SMA proteins in the lung tissues of mice in the high-and low-dose hesperidin groups significantly decreased(P<0.05).Compared with the those in the oe-NC group,the mice in the oe-EI24 group showed improved lung tissue morphology,less collagen deposition,and fibrosis formation;the inflammatory score and pulmonary fibrosis score,the expression levels of Collagen Ⅲ,Collagen I,Fibronection and α-SMA proteins in mouse lung tissu significantly decreased,and the expression levels of oe-EI24 and E-cadherin proteins significantly increased(P<0.05).Conclusion Hesperidin can improve the degree of pulmonary fibrosis in model mice,and its mechanism is related to the upregulation of E-cadherin expression and downregulation of Collagen I,Fibronection,and α-SMA expression.关键词
