Abstract

The purpose of this study is to analyze the residual elimination rule of carprofen injection in dairy cows and determine the withdrawal period.24 healthy dairy cows were administered carprofen at a dose of 1.4 mg/kg body weight via subcutaneous injection in the neck.Muscle,liver,fat and kidney samples were collected from cows at 12 h,3 d,7 d,14 d and 21 d time points after a single administration.After processing the tissue samples,the content of carprofen was detected by ultra-high performance liquid chromatography tandem mass spectrometry(UPLC-MS/MS).The methodological results showed that the method had a good linear relationship within the addtion range of 200~2000 ng/mL,and the correlation coefficient r≥0.99.The average recoveries of carprofen in different tissues were between 87.79%and 113.83%,the intra batch and inter batch coefficients of variation were less than 15%.Calculated by the calculation software WT1.4,the withdrawal periods of muscle,liver,kidney,and fat were 7.75 d,10.04 d,10.25 d,and 7.54 d,respectively.The residue of the injection site at 21 d was below the MRL in muscle.Therefore,carprofen injection should be used in dairy cows according to the recommended dosing regimen,and it is recommended to set the withdrawal period at 21 d.
