重复经颅磁刺激对脑缺血再灌注损伤大鼠及皮层小胶质细胞极化的影响
Effects of Repetitive Transcranial Magnetic Stimulation On Cerebral Ischemia-Reperfusion Injury and Polarization of Cortical Microglial in Rats

Abstract
Objective:To investigate the effect of repetitive transcranial magnetic stimulation on the polarization of cortical microglial in cerebral ischemia-reperfusion rats.Methods:SD rats were randomly divided into a sham operation group,a model group and an intervention group,with 8 rats in each group,and a rat model of cerebral ischemia/reperfusion injury was prepared by using middle cerebral artery lineal thrombus blockade.Rats in the intervention group received rTMS intervention 24 h after successful modeling,once a day for 7 days,and pseudo-rTMS stimulation was given to the sham-operation group and the model group.7 days later,all the rats were scored for neurological function and then sampled,and HE staining was used to observe the histopathological morphol-ogy of rat cerebral cortex,and the serum levels of IL-1β,TNF-α,IL-10,and TGF-β1 were detected by ELISA.In addition,fluores-cence quantitative PCR and Western blot were used to detect IL-1β,TNF-α,CD206 mRNA and protein expression in the cerebral cortex,respectively.Results:On the 2nd postoperative day,no neurological impairment was seen in the sham-operated group,and the neurological function scores of the rats in the model group were all significantly higher compared with the sham-operated group(P<0.0001);compared with the model group,the neurological function scores of the intervention group were elevated but statisti-cally indistinguishable(P>0.05).On the 8th postoperative day,no neurological impairment was observed in the sham-operated group,and the neurological function scores of the rats in the model group were significantly elevated compared with those in the sham-operated group(P<0.0001);the neurological function scores of the intervention group were significantly decreased compared with those of the model group(P<0.001).In the model group,the neurons were arranged in a disorganized way,some nerve cells died,and the nuclei of the cells were incomplete and difficult to recognize;in the intervention group,the degree of neuronal cell damage in the cerebral cortex and the number of reduced cells were reduced compared with that in the model group.Compared with the sham-operated group,the serum levels of IL-1β and TNF-α were significantly higher in the model group(P<0.001,P<0.0001),the levels of IL-10 and TGF-β1 were significantly lower(P<0.001),and the mRNA expression and protein levels of IL-1β and TNF-α in the cerebral cortex were significantly higher(P<0.0001,P<0.001),CD206 mRNA expression was significantly lower(P<0.0001)and CD206 protein level was significantly lower(P<0.05).Compared with the model group,serum IL-1β and TNF-α levels were sig-nificantly lower in the intervention group(P<0.05,P<0.001),TGF-β1 levels were significantly higher(P<0.01),IL-10 levels tended to be higher but there was no statistically significant difference(P>0.05),IL-1β and TNF-α mRNA expression in the cerebral cortex was significantly lower(P<0.001 P<0.05),IL-1β and TNF-α protein levels were significantly reduced(P<0.01),CD206 mRNA ex-pression was significantly elevated(P<0.01),and CD206 protein expression was elevated,but the difference was not statistically sig-nificant(P>0.05).Conclusion:Repetitive transcranial magnetic stimulation may alleviate cerebral ischemia-reperfusion injury in rats by modulating cortical microglial polarization to attenuate the inflammatory response.关键词
