摘要

Abstract

The objective of this study was to investigate the different ratios of antioxidant effects of daidzein(DA)and dihydromyricetin(DHM)combination in diets on ovarian granulosa cells(GCs)of laying hens and its effects on performance of laying hens during later laying period.The ovarian GCs were treated with tert-butyl hydroperoxide(TBHP)to establish an oxidative stress model,the cell experiment was divided into 5 groups:control group(group Ⅰ),TBHP group(group Ⅱ),TBHP+DA∶DHM(7∶3)group(group Ⅲ),TBHP+DA∶DHM(8∶2)group Ⅱ(group Ⅳ)and TBHP+DA∶DHM(9∶1)group Ⅲ(group Ⅴ).Animal experiment:a total of 192 Jingfen No.1 laying hens at 75 weeks of aged were divided into 2 groups according to similar egg production,with 6 replicates per group and 16 hens per replicate.The control group was fed a basal diet,while the experimental group was fed the basal diet supplemented with 300 mg/kg DA∶DHM(7∶3)combination.The pre-experimental period lasted for 1 week,and the experimental period lasted for 12 weeks.The results showed as follows:1)in the cell experiment,compared with group Ⅰ,the activities of su-peroxide dismutase(SOD),glutathione peroxidase(GSH-Px),catalase(CAT)and total antioxidant capacity(T-AOC)in GCs of group Ⅱ were significantly decreased(P<0.05),and the malondialdehyde(MDA)con-tent was significantly increased(P<0.05);compared with group Ⅱ,the activities of SOD,GSH-Px,CAT and T-AOC in GCs of groups Ⅲ,Ⅳ and Ⅴ were significantly increased(P<0.05),and the MDA content was significantly decreased(P<0.05).2)In animal experiment,compared with the control group,the egg production of experimental group was significantly increased(P<0.05),the feed-to-egg ratio was significantly decreased(P<0.05),the ovarian white follicle number and T-AOC were significantly increased(P<0.05),the ovarian MDA content was significantly decreased(P<0.01),the serum estradiol(E2)content was signifi-cantly decreased(P<0.05),and the contents of progesterone(P4),follicle stimulating hormone(FSH)and luteinizing hormone(LH)in serum were significantly increased(P<0.01).In conclusion,the combination addition of DA and DHM at a 7∶3 ratio can enhance the endogenous antioxidant enzyme activity in ovarian GCs of laying hens,increase the ovarian antioxidant capacity of laying hens during the later laying period,reduce MDA content,and increase serum P4 and FSH contents,thereby improving the performance of laying hens in the later laying period.[Chinese Journal of Animal Nutrition,2025,37(2):1022-1032]
