Protective Effects of Angelica sinensis by-Product on Pellet Feed and Its Effects on Growth and Hypoxic Stress in Crucian Carp

Abstract
The purpose of this study was to investigate the protective roles of Angelica sinensis by-product(AP)on pellet feed and its effects on growth and hypoxia stress in fish.Firstly,seven experimental diets were formulated by adding AP to the basal diet to provide a final concentration of 0(control),1.0%,2.0%,4.0%,8.0%,16.0%and 32.0%,respectively,and make it into 1.5 mm pellet feed.Then,420 crucian carp[Carassais auratus gibebio,body weight of(5.67±0.07)g]were selected and randomly divided into 7 groups,with 3 replicates in each group and 20 fish in each replicate.Seven groups were fed with diets contai-ning different levels of AP for 60 days.The results showed that AP at the addition level of 16.0%increased the moisture content(P<0.05),and at the addition levels of 4.0%to 16.0%,1.0%to 8.0%and 8.0%to 32.0%reduced hardness,powder rate and mildew growth in pellet feed,respectively(P<0.05).Dietary AP at the addition levels of 2.0%to 8.0%,2.0%to 8.0%,8.0%to 16.0%,8.0%to 32.0%,8.0%,2.0%to 16.0%,1.0%to 16.0%,8.0%to 32.0%,2.0%to 32.0%,4.0%to 32.0%,2.0%to 8.0%,8.0%,1.0%to 32.0%and 2.0%to 32.0%increased the weight gain rate(WGR),special growth rate,feed efficiency,durative time(DT)after hypoxia,hematocrit,red blood cell count and hemoglobin content in the blood,the activities of superoxide dismutase(SOD)and catalase and the content of reduced glutathione(GSH)in red blood cells and the activities of SOD,glutathione peroxidase and glutathione reductase and the content of GSH in gills(P<0.05),respectively.Meanwhile,dietary AP at the addition levels of 8.0%to 32.0%,1.0%to 16.0%,8.0%,4.0%to 8.0%,2.0%to 32.0%,4.0%to 16.0%,2.0%to 32.0%and 4.0%to 32.0%reduced the oxygen consumption rate(OCR)after hypoxia,mean corpuscular volume in the blood,the levels of superoxide anions(O2),hydrogen peroxide and methemoglobin in red blood cells as well as O2-,hydroxyl radicals and malondi-aldehyde in gills of crucian carp(P<0.05),respectively.These results indicate that AP improve the solidity and has anti-mold and water retention effects in pellet feed.Dietary AP increase the growth performance,hy-poxia tolerance,and antioxidant capacity in gills and red blood cells of fish.According to the broken-line re-gression analysis based on WGR,the appropriate and highest addition levels of AP are estimated to be 8.08%and 23.07%in diets,respectively.The broken-line regression analysis based on DT and OCR show that the ap-propriate addition levels of AP to improve hypoxia tolerance in fish are 7.62%and 7.85%in diets.The im-provement of hypoxia tolerance in fish by AP may be closely related to its reduction of aerobic metabolism and increase in the number of red blood cells and Hb content in blood as well as the antioxidant capacity in gills and red blood cells of fish.In summary,AP has a protective effect on pellet feed and can improve the growth and hypoxia tolerance in fish.[Chinese Journal of Animal Nutrition,2025,37(2):1173-1187]关键词
