响应面法优化艾蒿多糖的提取工艺及其组成成分和体外抗氧化活性分析
Optimization of Extraction Process of Artemisia argyi Polysaccharide by Response Surface Methodology and Analysis of Its Components and in Vitro Antioxidant Activity

Abstract
This experiment was conducted to optimize the extraction process of Artemisia argyi polysaccharides(AAP),and investigated it components and in vitro antioxidant activity.Based on single-factor experiments,the extraction yield of AAP was taken as the response value,and the optimal extraction conditions were deter-mined through response surface methodology.The shape,structure,monosaccharide composition and molecu-lar weight of the purified AAP were analyzed.Additionally,its antioxidant activity of AAP was evaluated by measuring the scavenging rates of 1,l-diphenyl-2-picrylhydrazyl radicals(DPPH·),hydroxyl radicals(·OH)and superoxide anion radicals(O2-·)as well as its iron reducing power.The results showed as fol-lows:1)the best extraction process of AAP was optimized by response surface with liquid to material ratio of 35:1,enzymatic hydrolysis time of 4 h and temperature of 50 ℃.The extraction rate of AAP was 12.46%,and the content of soluble polysaccharides was 297.11 mg/g.2)The purified AAP exhibited a variety of irreg-ular particle shapes,including rough cubic particles,long-chain flakes and spherical forms.It contained five monosaccharides:arabinose,galactose,glucose,xylose and mannose,with molecular weight ratios of 1.68:3.54:80.81:2.38:11.59.The average molecular weight was 114.786 ku,and it was classified as a pyranose.3)At the same concentration,the DPPH·scavenging rate and iron reduction rate at concentrations ranging from 0.05 to 1.0 mg/mL in the AAP group were significantly lower than those in the positive control group(P<0.01),and the·OH scavenging rate at concentrations ranging from 0.40 to 1.00 mg/mL and the O2·scavenging rate at concentrations ranging from 0.20 to 1.00 mg/mL were also significantly lower than those in the positive control group(P<0.01).The half-maximal inhibitory concentrations(IC50)for DPPH·,·OH,and O2-·scavenging rates were 0.055 2,0.868 4 and 0.839 2 mg/mL,respectively.In conclusion,this study optimizes the traditional AAP extraction process and demonstrates that AAP exhibits robust antioxidant activity through component analysis and in vitro antioxidant activity determination.This provides a robust theoretical foundation for the development and utilization of AAP as a feed additive.[Chinese Journal of Animal Nutri-tion,2025,37(2):1271-1285]关键词
