槲皮素对小鼠肌内脂肪沉积的影响及其机制探究
Effects of Quercetin on Intramuscular Fat Deposition in Mice and Its Mechanisms

Abstract
The experiment was conducted to investigate the effects of quercetin on intramuscular fat(IMF)deposition in mice and its potential regulatory mechanisms.Twenty-four 7-week-old male C57BL/6J mice were selected and randomly divided into two groups as control group(NCD group)and quercetin group(NQ group)with 6 replicates in each group and 2 mice in each replicate.Mice in NCD group were gavaged with 10 μL/g BW phosphate buffer solution(PBS)daily,while those in NQ group were gavaged with 10 μL/g BW quercetin solution(10 mg/mL)daily.The experimental period lasted for 42 days,during which all mice were freely fed a basic diet and drinking water.On the 29th day of the experiment,50%glycerol was injected into the bilateral tibialis anterior of mice to induce the establishment of muscle fat infiltration model,and mus-cle samples were taken on the 7th and 14th day after the modeling.Male C57BL/6J mice aged at 8 weeks were selected for cell experiment,and mouse fibro/adipogenic progenitors(FAPs)were isolated and cultured.Cocktail method was used to induce adipogenic differentiation while treating with gradient quercetin(0,1,5,10,20 and 40 μmol/L).The results showed as follows:1)100 mg/kg BW quercetin inhibited IMF deposi-tion in mice,but had no significant differences on mice weight,feed intake,and serum triglyceride content(P>0.05).2)Quercetin dose-dependently inhibited FAPs differentiation polyester.3)Quercetin altered the fe-cal microbiota structure of mice,reduced the relative abundances of Firmicutes and Bacteroidota as well as Fir-micutes to Bacteroidota ratio,while increased the relative abundances of Romboutsia and Ileibacterium.4)Quercetin altered the fecal metabolites of mice,and the differential metabolites were enriched in pathways relat-ed to amino acid,fatty acid and bile acid metabolism.Among them,compared with NCD group,the fecal contents of metabolites such as spermine,spermidine and inosine in NQ group were significantly increased(P<0.05),while the fecal contents of metabolites such as eicosapentaenoic acid(EPA)and 20-hydroxyeicosatet-raenoic acid(20-HETE)were significantly reduced(P<0.01).5)Quercetin significantly increased the mR-NA N6-methyladenosine(m6A)level in tibialis anterior of mice(P<0.05).In summary,quercetin can inhibit IMF deposition in mice,which may exert regulatory effects through multiple pathways such as affecting the structure of mice gut microbiota and related metabolites and muscle tissue m6A methylation modification.[Chi-nese Journal of Animal Nutrition,2025,37(2):1298-1311]关键词
